No. Not unless you had earlier calibrated with an object closer to the pole than to the celestial Equator. Otherwise the calibration should be good for at least the entire night.
If you had calibrated near declination = 0, then the calibration should be accurate enough to use everywhere in the sky. You also need to flip the calibration after doing a Meridian Flip, but that only involves changing the sign of the calibration, and does not need a full recalibration.
In general, you only need to re-do calibration if you have changed the camera, adjusted the direction of the guide scope, the guide camera angle, and/or plate scale of your guide scope, or the guiding settings (in the Guide setup window).
